When i change the layout in the post page settings. The posts and Ads in the right column get auto removed. What is the issue.

This is no issue, you chose a standard template in your image, and the standard template uses for sidebar widgets, those can be set from the appearance widgets. The single post template is a cloud template https://forum.tagdiv.com/how-use-tagdiv-cloud-library-templates/ that can be edited on frontend using tagDiv Composer and you can create the layout as you like https://forum.tagdiv.com/tagdiv-composer-tutorial/

How to apply post template to all the posts in a single shot instead of doing individually to east post page.

By default, the post template is set to get on from template – https://i.imgur.com/kA1wmeR.png and it will get the post template set in the theme panel > post settings > default post template – https://i.imgur.com/P0UvELX.png
