Can we enable AMP without using mobile theme plugin, so that we can even customize amp pages according to our choice which is impossible if we have mobile theme plugin active in website

Yes, you can use the amp plugin with out mobile theme, use the option like this -> https://i.imgur.com/lbe43pJ.png but the best implementation of this amp with our theme is using it with mobile theme plugin.
