How to change the post with big img to the right side? and the post lists to the left side?

The theme doesn’t have an option to change the post displayed in blocks individually, but you can apply different filters like categories, authors, tags from block settings: http://screencast.com/t/waB0NSOVcZ5

Note that I’ve test this only with a 2/3 + 1/3 layout: http://screencast.com/t/Vt549TmPWL and only with block 1. For a different implementation you will need custom modification and if you’re not aware of the steps you need to take in this regard please consider hiring a developer or a freelancer to do this for you.
