I have set the Adsense size to 640×300 for Header. But no effect. Still the ad size is small in height.
Also, can I use size 970 x 250 size on header.

Please make sure you clear cache after applying changs. The ad size is selected but no ad can be delivered in that size:https://www.screencast.com/t/3PtS7Nw9YI8w The contianer of the header will not permit it.

you can try to tweak it with css a bit but the ad is way too big for the header ad spot. It will not look good. There is no simple solution to implement both that huge ad and the logo on the same row. I this case i recommend using a header style that does not display the header ad next to the logo.

The header styles that display the logo and the header ad on the same line, these would require contain a 728 x 90 ad, that would be the proper size
